Amyloid cascade
Model proposed in 1992 whereby the accumulation of amyloid beta peptides in the brain triggers tau hyperphosphorylation, neuroinflammation and the cognitive decline of Alzheimer's disease. Still the dominant framework of the 2024 Alzheimer's Association diagnostic criteria, but nuanced by largely disappointing anti-amyloid trials and by non-linear patient trajectories.
